Preparation and Evaluation of the Release Kinetics of Diltiazem Hydrochloride from Tableted Microspheres

Abstract

Microsphere is one of the multiple unit dosage forms.  microspheres were prepared by solvent evaporation method. Diltiazem hydrochloride and each polymer mixture were dissolved completely in acetone-methanol mixture by stirring at 500 rpm  in ice-bath at 10oC for 10 minute.  Above mixture was poured rapidly into the liquid paraffin previously cooled at 10oC, while it was being stirred by mechanical stirrer at 1000 rpm.  Resulting emulsion was agitated at 35oC for 4 hours and the organic solvent, acetone-methanol were removed completely by evaporation. Solidified microspheres were filtered through Whatmann filter paper. Scanning of Diltiazem hydrochloride was performed into Methanol, acid buffer (pH 1.2) and phosphate buffer (pH 7.2).  The characteristic peak, lmax was found to be at 237, 237 and 239 nm respectively. The entrapment efficiencies ranged from 86.11 – 98.90%, Drug release from microspheres was determined using paddle type in-vitro dissolution method. This slow increase in the release behaviour was observed in the formulation F5, F6, F7, F8, F13, F14, F15 and F16; rapid increase in release behaviour was observed in the formulation F1, F3, F9 and F11. Whereas almost the linear behaviour after the burst release was observed in the formulations F2, F3, F9, F10, F12.
